Gene Orders Overview

1: Enter the name of your Gene
2: Choose your Sequence Type (DNA or Protein) - Default is DNA
3: Choose your service of choice (Gene Synthesis, Subcloning Only or Mutagenesis) - Default is Gene Synthesis

4: Paste your sequence in the text box
5: If there are any invalid characters in your sequence, you can click the "Show" button to see the characters
note: You can also see the length of the DNA bases

6: Invalid characters will be highlighted in red, please correct them in your sequence above

7: Select "Yes" if you would like to have your sequence optimized - Default is "No"
7.1: If "Yes," please indicate the specific species you would like your gene optimized for expression within (for e.g. E.Coli, Homo sapiens, Mus musculus, Nicotiana benthamiana, etc)
7.2: Please enter any restriction sites you would like us to avoid/maintain within your sequence (for e.g. Maintain-XhoI, NotI, BamHI Avoid-EcoRI, SpeI, EcoRV)
8: Select "Yes" if a stop codon is required - Default is "No"

9: Have you entered a Kozak sequence in the textbox above? If so, please select "Yes" - Default is "No"
9.1: If "Yes," please also include the sequence in this text box.
note: Please inlude the Kozak sequence in both the main "Sequence" text box as well as the "Kozak Sequence" text box, or your quote will be innacurate

10: Please select your vector of choice from the dropdown menu - Default is pUC57-Amp
10.1: If you would like to submit your own vector, please select "OTHER - User Submitted" from the dropdown menu
note: Please provide additional details if you submit your own vector (such as vector name, antibiotic resistance and growth conditions)

11: Indicate the restriction sites present within your sequence to be used for cloning (for e.g. 5' Site: NotI, 3' Site: XhoI) - Default is blunt-end cloning
12: Select the delivery amount of your gene from the dropdown - Default is 2-4 ug
13: Choose additional deliverable types, note that you will always be provided a lyophilized gene in addition to other type(s) you select
note: Blunt-end cloning is only available for pUC and Bluescript vectors

14: Click on "Calculate Price" to see your Gene Order Summary and Price

15: You can now view your order summary for genes or go back to make any edits
16: Click on "Checkout" to proceed
note: Pricing on this page is an estimate and does not include further discounts, tax or shipping. You will see this in the final order summary

17: Please select your store for checkout (Canada if you are located in Canada, or United States if you are located in USA respectively)

You can now view your genes in the Bio Basic Shopping Cart
18: Please apply any any promo code here (if available). You can also view the tax and shipping prices by clicking on "Estimate"
19: Click on "Proceed to Checkout" to see the Final Summary with all the pricings included

20: After filling in your Billing, Shipping and Payment information, you can click on "Place Order" begin processing of your gene by our gene team

Uploading Genes in Bulk

1: Click on "Bulk Gene Import"
2: Download the appropriate template file(s) based on your desired service (Gene Synthesis, Subcloning or Mutagenesis)

3: In the template, please filled in the required fields at minimum. One row per gene.
4: Please fill in the additional fields if applicable to your gene(s). If blank, the default is 2-4ug gene blunt-end cloned in a pUC57-Amp vector
note: Be sure to save your filled template file

5: Click on "Choose File" to attach your completed template file(s)
6: Click "Import" to import your genes from the file(s) into the portal. You can import template files one at a time
note: To avoid duplicate gene entries, please do not import the same file more than once (unless desired)

7: After you import your genes, you can view any respective errors (*: invalid gene sequences)
8: Please review and fix any invalid gene sequences (if appliable), then click on "Calculate Price" to proceed
note: Since you are directly correcting errors on the portal, invalid sequences would still remain in your template file, unless you edit the file itself

9: Review the summary of your genes, then click on "Checkout" to add your genes to your shopping cart

Using Protein Sequences

1: Enter the name of your gene
2: Select the Sequence Type as "Protein"
3: Enter the Protein Sequence as single letter amino acid sequences in the large text box
4: Enter flanking DNA Sequences in the two smaller text boxes if applicable

Submitting your own vectors

1: Select "OTHER - User Submitted" from the drop-down vector list
2: Enter additional information on the vector such as vector name, antibiotic resistance and growth conditions as available
note: You can also add attachments as PDF, Excel, Word, etc.

To send your own vectors:
Please send two tubes each containing 4.0 ug of plasmid DNA to:

Subcloning-Only Orders

1: Select "Subcloning Only" as the service type
2: Enter the Final DNA sequence of the gene (without the Final Plasmid DNA sequence)
3: Enter the Source DNA sequence of the gene (with the Source Plasmid DNA sequence)
4: Enter the Final DNA sequence of the gene (with the Final Plasmid DNA sequence)
5: Select the vector, cloning sites, delivery amount and deliverable type(s)

1: Click on "Proceed to Checkout" on the Shopping Cart page to go to the Checkout Page
